چکیده

The sedimentation and diffusion coefficients have been determined for Hemophilus influenzae transforming activity and DNA using P(32)-labeled DNA. The methods employed the Spinco fixed boundary separation cell for measurements of the sedimentation coefficient and the Northrop-Anson diffusion cell to determine the diffusion coefficient. There was a very close correlation between the amount of DNA and transforming activity sedimented or diffused. The sedimentation coefficient (s(20 degrees )), for both biological activity and DNA was 27 and the diffusion coefficient (D(20 degrees )) 1 x 10(-8) cm(2)/sec. The molecular weight calculated from these coefficients gave a value of 16 million. There was no difference in the sedimentation coefficients for the two unlinked markers, streptomycin and erythromycin resistance, and the diffusion coefficients for single markers or the linked markers, streptomycin and cathomycin, were the same.

Comparison of two nonculture techniques for detection of Hemophilus influenzae in sputum. In situ hybridization and immunoperoxidase staining with monoclonal antibodies.

Two nonculture methods, in situ hybridization and immunoperoxidase staining with monoclonal antibodies, were compared for the detection of Hemophilus influenzae in 184 sputa. For in situ hybridization, a biotin-labeled probe of total genomic DNA of H influenzae type b was prepared that hybridizes specifically with H influenzae, H parainfluenzae, H hemolyticus, and H parahemolyticus DNA. Immunop...

Vaccine Candidates against Nontypeable Haemophilus influenzae: a Review

Nonencapsulated, nontypeable Hemophilus influenzae (NTHi) remains an important cause of acute otitis and respiratory diseases in children and adults. NTHi bacteria are one of the major causes of respiratory tract infections, including acute otitis media, cystic fibrosis, and community-acquired pneumonia among children, especially in developing countries. The bacteria can also cause chronic dise...

Plasmid transformation in Haemophilus influenzae.

Purified 34-megadalton-plasmid deoxyribonucleic acid from antibiotic-resistant strains of Haemophilus influenzae transforms competent strains of H. influenzae more efficiently if the recipient strains contain certain other 30-megadalton plasmids.

Antigenic heterogeneity of lipid A of Haemophilus influenzae.

The chemical structure and biologic function of the lipid A portion of lipopolysaccharide are not identical among gram-negative bacteria. This study indicates that antigenically heterogeneous lipid A exists among strains of Haemophilus influenzae. An immunoglobulin G3 murine monoclonal antibody, 3D2, produced against a nontypable H. influenzae strain 3524 has specificity for a site on the lipid...

Attempts to demonstrate type transformation with Klebsiella pneumoniae.

Since Griffith's demonstration of capsular type transformation in pneumococci a vast amount of research has been done on pneumococcal and other type transformations with a variety of genetic markers. (For general reviews see Ephrusi-Taylor, 1951; Austrian, 1952.) Type transformations in pneumococci have been obtained by in vivo and in vitro methods. For in vitro transformations competent cells,...
